Method Development for the Separation of Phospholipids by Subcritical Fluid Chromatography

چکیده
Super/subcritical Fluid Chromatography (SFC) has been used successfully for the analysis of a variety of compounds including foods, polymers, and pharmaceuticals. 1-7 Supercritical carbon dioxide exhibits solvating power comparable to hexane and has been primarily utilized as a fluid for the separation of relatively non-polar compounds. 8 For this reason, many SFC methods have been developed as substitutes for normal-phase HPLC methods. Commonly used normal-phase HPLC mobile phases consist generally of chlorinated solvents such as chloroform and methylene chloride. Traces of water in these mobile phases cause retention times to fluctuate in the normal-phase mode. These solvents also present an environmental hazard, which can be eliminated in part by using a mobile phase of carbon dioxide which may or may not have been modified with a small percentage of organic solvent. Additionally, the quantity of solvent required and the high disposal costs commonly encountered with many normal-phase HPLC solvents may be dramatically reduced by employing carbon dioxide based mobile phases.

Subcritical water extraction (SWE) of essential oil from Matricaria Chamomilla L. (M. Chamomilla L.) was performed and compared with conventional method including hydro distillation. The percentage of main extracted components of different temperatures (100, 125, 150 and 175 oc) and water flow rates (1, 2 and 4 ml/min) were investigated at 20 bar pressure and 0.50 mm mean particle size in SWE....
متن کاملSimulation of Separation of a Racemic Mixture of Ibuprofen by Supercritical Fluid Chromatography in Simulated Moving Bed
Separation of a racemic mixture of ibuprofen at a low concentration level by supercritical fluid chromatography in a simulated moving bed (SFC-SMB) is investigated by simulation. The feasibility of ibuprofen enantiomers separation has been experimentally examined in the literature. Our simulation results show that separation of ibuprofen enantiomers is feasible by this method, and R-ibuprofen a...

Background: Fatty acids are known to be critically important in multiple biological functions. Phospholipid fatty acids of follicular fluid, an important microenvironment for the development of oocytes, may contribute to the women’s fertility and the efficacy of assisted reproduction techniques. The aim of this study was to investigate the effect of fatty acid composition of follicular fluid ph...
متن کاملAmniotic fluid phospholipids measured by continuous-development thin-layer chromatography.
We describe a one-dimensional thin-layer chromatographic system for separation of amniotic fluid phosphatidylcholine, sphingomyelin, phosphatidylglycerol, phosphatidylinositol, phosphatidylserine, and phosphatidylethanolamine in amniotic fluid. We utilize short-bed continuous development and "high performance" thin-layer chromatography. Phospholipids are detected with an antimony molybdate stai...
متن کاملDevelopment of a new method based on chiral ligand-exchange chromatography for the enantioseparation of propranolol
A new chromatographic procedure was proposed for the separation of propranolol (PRN) enantiomers based upon enantioselective chiral ligand-exchange chromatography. The separation was carried out on a short C8 column leading to considerably short separation time. L-alanine and Cu2+ were applied as chiral selector and central bivalent complexing ion, respectively. It was found that the kind of co...
